For most customers, we recommend performing initial testing with shRNAs in a viral vector backbone (i.e., transfect with the shRNA plasmids and validate in vitro). We guarantee at least 70% knockdown of your gene of interest with Vigene-designed shRNA vectors. Once you have determined the best shRNA for your experiments, we can then package your validated AAV vector for efficient gene knockdown in vivo.

Pre-designed shRNA Vectors

Our shRNA clones are provided as a set of 4 shRNA vectors in the pAV-U6-GFP backbone (default vector) and a free scrambled shRNA vector control. The constructs can be transfected as is or can be packaged into virus for efficient gene delivery.

What is Vigene’s guarantee for shRNA cloning services?
We guarantee that at least one shRNA construct in the set of four will yield at least 70% knockdown of target gene expression as determined by qRT-PCR. If you do not achieve this level of knockdown, Vigene will replace the construct(s) one time at no additional cost.
Does Vigene validate shRNA sequences?
Vigene does not validate shRNA sequences designed as a part of the shRNA cloning services. We will send you the shRNA plasmids to test. Once you determine which shRNA construct works best for your experiments, we can package your selected shRNA construct into virus.
