How do I Know Which AAV Control to Use?

There are three major variables to consider when choosing an appropriate control: the promoter, the reporter, and the serotype. The promoter should ideally match that of the virus expressing your GOI. However, you have some flexibility with the reporter you choose as it will depend on your experimental conditions. Many scientists use fluorescent protein reporters, such as GFP AAV or RFP AAV, for their ease of use and detection. Lastly, the AAV serotype should match the tissue where your GOI is expressed. If you are unsure which promoter or serotype will work best for your experiments, please see the FAQ section at the bottom of this page.

What is Vigene’s guarantee?
Vigene guarantees that our AAV controls will contain the specified sequences detailed on their product description pages and specified viral titer. Please note that we cannot guarantee expression because it will depend on your experimental conditions. An AAV control with a tissue-specific promoter may only express in the intended tissue.

Which serotype should I use?

Specificity is determined by the viral capsid proteins, so it is important to select the correct AAV serotype to ensure optimal gene delivery. The table below lists our recommended AAV serotypes for different target tissues. If you are unsure which AAV serotype will work best for your system, we recommend that you try out our AAV GFP Testing Kit. The testing kit contains the following viruses all driven by the strong CMV promoter: GFP AAV1, GFP AAV2, GFP AAV5, GFP AAV6, GFP AAV8, GFP AAV9, and GFP AAV-DJ.
