What Types of AAV Controls Are Available?
AAV Empty Capsids - New Products
We are offering AAV empty capsids in serotypes AAV1, AAV2, AAV5, AAV6, AAV8, and AAV9 (see the products in the table above). The AAV empty capsids are produced and purified using iodixanol gradient ultracentrifugation. The final quality control assays include ELISA for titering, endotoxin assay, bioburden, SDS-PAGE and silver staining for purity.
AAV Reporter Controls – AAV Controls Expressing Reporters
Vigene provides >2000 AAV controls expressing a variety of reporters driven by many different type of promoters, constitutive and inducible systems. The reporters include GFP, mCherry, RFP, LacZ, and Luciferase. The AAV reporter controls are available in AAV1, AAV2, AAV5, AAV6, AAV7, AAV8, AAV9 and AAV0DJ.
How do I Know Which AAV Control, Reporter Control to Use?
There are three major variables to consider when choosing an appropriate AAV reporter control: the promoter, the reporter, and the serotype. The promoter should ideally match that of the virus expressing your GOI. However, you have some flexibility with the reporter you choose as it will depend on your experimental conditions. Many scientists use fluorescent protein reporters, such as GFP AAV or RFP AAV, for their ease of use and detection. Lastly, the AAV serotype should match the tissue where your GOI is expressed. Which serotype should I use?
Specificity is determined by the viral capsid proteins, so it is important to select the correct AAV serotype to ensure optimal gene delivery. The table below lists our recommended AAV serotypes for different target tissues. If you are unsure which AAV serotype will work best for your system, we recommend that you try out our AAV GFP Testing Kit. The testing kit contains the following viruses all driven by the strong CMV promoter: GFP AAV1, GFP AAV2, GFP AAV5, GFP AAV6, GFP AAV8, GFP AAV9, and GFP AAV-DJ.
